"PNAS": Academician Shi Yigong analyzes key proteins of AD

Alzheimer's disease (AD) is the most common form of dementia, but we still know very little about the etiology of AD. Recently, researchers from Tsinghua University tested the effects of 128 AD-derived Presenilin-1 (PS1) mutations on the production of amyloid β (Aβ42 and Aβ40) using highly purified recombinant γ-secretase. The findings were published in the December 5th issue of the Proceedings of the National Academy of Sciences (PNAS).

Leading the study is Professor Yigong Shi of Tsinghua University. Shi Yigong's research group focuses on the molecular mechanisms of tumorigenesis and apoptosis using structural biology and biochemistry, focusing on the structural and functional studies of tumor suppressor and apoptosis-regulating proteins, and major disease-associated membrane proteins. Research on structure and function, structure and function of intracellular biological macromolecular machines. After returning to China, Shi Yigong published several papers in international top journals such as Nature. At the same time, he also built a talent introduction bridge centered on Tsinghua University. In 2013, Shi Yigong was elected as an academician of the Chinese Academy of Sciences.

Shi Yigong

In 2016, Shi Yigong's research team achieved a series of research results. In the beginning of the year, he published a heavy result in Science magazine. They obtained U4/U6.U5 three small nuclear ribonucleotides with a resolution of 3.8 angstroms. The three-dimensional structure of the protein complex (U4/U6.U5 tri-snRNP) provides new insights into the assembly and catalysis of spliceosomes. In July, two research papers were published in the same issue of Science, and the atomic structure of the catalytically activated splice (Bact) from Saccharomyces cerevisiae was determined by cryo-electron (cryo-EM) and averaged. S. cerevisiae with a resolution of 3.4 angstroms catalyzes the atomic structure of the first-stage splicing complex (complex C). In addition, in February and March of this year, Academician Shi Yigong also published as a correspondent in PNAS. Two research results.

One hallmark of Alzheimer's disease (AD) is the aggregation of beta-amyloid (Aβ) in the brain of a patient into amyloid plaques. Amyloid precursor protein (APP) is cleaved by intramembrane protease gamma secretase, resulting in varying lengths of A[beta], with longer peptides (such as [beta]42) being considered detrimental. Increasing the ratio of longer Aβs beyond short Aβs, for example, increasing the ratio of β42 over Aβ40, may lead to the formation of amyloid plaques and consequent development of AD.
