The team of Professor Xia Ning Shao from Xiamen University and the team of Professor Li Jun from the First Affiliated Hospital of Zhejiang University have developed a first high-simulation mouse model of human liver cirrhosis caused by hepatitis B virus in the world after five years of collaborative research. Simulating the whole process of human chronic hepatitis B cirrhosis induced by natural infection of hepatitis B virus (HBV), solving the international problem of HBV infection and pathogenesis research and the lack of high-quality animal models for the development of new drugs against hepatitis B cirrhosis. The research results were published online on January 30th in the international authoritative journal Gastroenterology, Gut.

At present, there are nearly 250 million patients with chronic hepatitis B caused by HBV infection, of which 80 million are in China, and 600,000 people die of HBV-related diseases every year, which is a serious threat to human health. And so far there is no high simulation of HBV infection caused by the animal model of slow hepatitis B and cirrhosis, which restricts the basic research and new drug development related to HBV.

Researchers from Xiamen University and Zhejiang University first constructed a model of fulminant hepatic failure using immunodeficient FRGS mice, followed by transplantation of human bone marrow mesenchymal stem cells (BMSCs), and finally regenerating human liver cells and various immunitys in the liver of mice. Cells, successfully constructed a mouse model of hepatocyte and immune cell double-sourced. After the mouse model is infected with HBV, the key indicators of HBV continue to be positive, long-term chronic inflammation, and progressive liver fibrosis and cirrhosis. This is the first time that a single human BMSC transplantation has been used to realize the dualization of human hepatocytes and various immune cells in mice, and to simulate the natural infection of human HBV and the pathogenesis of hepatitis B cirrhosis.

The animal model of disease is the cornerstone of the development of modern medicine, especially in the case of major and sudden outbreaks of infectious diseases. The appropriate animal model of disease can provide a strong guarantee for the timely detection of pathogens and the development of prevention and control strategies. The original disease animal model has become a measure of a country. The mark of the level of biomedical research. This move means that China is currently at the international leading level in the field of animal models of hepatitis B cirrhosis.
