Analyze the metastatic potential of circulating tumor cells

Circulating Tumor Cell (CTC) is a general term for various types of tumor cells existing in peripheral blood. It is detached from solid tumor lesions (primary lesions and metastases) due to spontaneous or diagnostic operations, and most CTCs enter peripheral blood. After apoptosis or phagocytosis, a few can escape and anchor to develop metastases, increasing the risk of death in patients with malignant tumors.

Numerous studies have shown that CTC exists in peripheral blood in different forms, ranging from free single CTC to aggregated cell cluster (CTM). Epithelial-mesenchymal transition (EMT) occurs in tumor cells as they enter the peripheral blood circulation. Therefore, there are different types of CTC, including epithelial cell phenotype, interstitial cell phenotype, and mixed phenotype of epithelial cells and interstitial cells. CTM and the mesenchymal cell phenotype CTC have stronger metastatic potential.

Studying tumor biology through CTCs - a renaissance caused by technological advances!

Circulating tumor cell CTCs, the Australian pathologist John Ashworth first proposed this concept in 1869. Studying the rare and rare tumor cells in a background condition of about 5 million monocytes in 1 ml of blood is like a needle in a hayst, which is the main technical challenge facing CTCs research. Today, superior technological developments in cell enrichment, separation, molecular analysis, and bioinformatics reignite research in the field of CTCs. Studies have found that the spread of tumor cells is the leading cause of death in cancer patients.

The enrichment of CTCs, such as cell size and morphology, is exploited by the physical characteristics of tumor cell surface markers or tumor cells; these are characteristic of tumor cells. Any technology that enriches CTC may have the same characteristics of rare tumor cells and blood cells, and cannot be enriched in the true sense of circulating tumor cells.

A recent treatment technique completely skips the step of circulating tumor cell enrichment, but identifies each cell, followed by the identification of tumor cells, through sophisticated single-cell selection devices, or through dielectrics. Physical separation method for charge transfer single-CTC molecular analysis.

The patient's circulating tumor cells can grow in vitro or in immunocompromised mice, and their biological behavior is observed by high-resolution microscopy.

Circulating tumor micro-embolism - the gang's collaborative survival journey!

The formation of circulating tumor micro-plugs (CTM), the cluster of CTCs, is the most important threat to tumor metastasis. CTM cell components include platelets, stromal cells, hematopoietic cells, etc.; however, the role of each cell in CTM is unclear. The prevailing view is that CTC alone is prone to death during the cycle, while tumor cells in tumor microtubules are protected from apoptosis. How a CTM containing 20-30 cells forms new lesions through narrow capillaries, perhaps to enhance the protection of tumor cells from immune system attacks.
